摘要:
以西藏帕里产粗壮秦艽为研究对象,采用硅胶柱、Sephadex LH-20凝胶柱等手段进行化学成分的分离纯化,通过波谱数据(MS,1H-NMR,13C-NMR)及标准品对照进行结构鉴定;进一步利用HPLC法测定该植物不同器官(根、茎、叶、花)中几种成分的含量并比较含量差异. 从西藏帕里产粗壮秦艽中分离得到5个化合物,分别鉴定为栎瘿酸(1)、谷甾醇(2)、龙胆苦苷(3)、马钱苷酸(4)、獐牙菜苦苷(5),其中环烯醚萜苷类化合物马钱苷酸、獐牙菜苦苷、龙胆苦苷、獐牙菜苷的含量较高,且在4种器官中含量差异明显. 本研究首次从西藏帕里产粗壮秦艽中分离得到化合物1-5,其中环烯醚萜苷类含量丰富,在4种器官中马钱苷酸和龙胆苦苷总含量均达到2015年版药典标准,因此根、茎、叶与花均具有同等重要的开发价值. Abstract:
The aim of this paper was to systematically study the chemical constituents of Gentiana robusta from Tibet, China. The constituents were separated by silica gel and Sephadex LH-20 column chromatography. Then, their structures were identified on the basis of spectral data (MS, 1H-NMR, and 13C-NMR) or by comparison with standard compounds. Four iridoid glucosides including loganic acid, swertiamarin, gentiopicroside, and sweroside in four organs (root, stem, leaf, and flower) of G. robusta were identified by HPLC, and concentrations in different organs were compared. Five compounds were isolated from G. robusta in Tibet and their structures were identified as roburic acid (1), β-sitosterol (2), gentiopicroside (3), loganic acid (4), and swertiamarin (5). The identification results showed that loganic acid, swertiamarin, gentiopicroside, and sweroside were abundant in G. robusta, and the concentrations in different organs varied. Compounds 1–5 were isolated from this species for the first time. It appears that concentrations of four iridoid glucosides are high in four organs of G. robusta, and the summed amounts of gentiopicroside and loganic acid in the root, stem, leaves, and flowers are up to the standards of Chinese Pharmacopoeia (2015). The root, stem, leaves, and flowers have equal development values.
